Molecular genetic studies on the mode of action of azole antifungal agents.

چکیده
Cytochrome P-450 catalysing the 14ademethylation of sterol is inhibited in fungi by azole antifungal compounds resulting in a fungistatic or a fungicidal effect [ 11. The azoles themselves represent a useful tool for investigating P-450 structure through the differential affinities with different P450 forms. [Z]. Of practical importance is the identification of the molecular features which result in the strong inhibition of a cytochrome P-450 and which may lead to the development of better antifungals, as well as other therapeutic agents and agrochemicals. The extensive use of azoles is also resulting in increasing reports of resistance in agriculture and in the clinic and we have been interested in the relationship of the resistance mechanisms to the target enzyme and the mode of action of azole antifungals

Design, Molecular Docking Studies and Toxicity Prediction of Some Novel 1, 2, 3-Triazole Derivatives Containing Piperazine Moiety as Antifungal Agents and CYP-51 Inhibitors
Background & Objective: In this study, a number of new triazole derivatives, containing a 1, 2, 3-triazole ring attached to the piperazine moiety as antifungal agents and lanosterol 14 alpha-demethylase, (CYP51) inhibitors were docking studies conducted. In the following, the toxicity risks of the designed compounds, were predicted by existing software. Materials & methods: Initially, the chemi...
متن کاملAntifungal agents: Polyene, azole, antimetabolite, other and future agents
Antifungals have always been considered as one of the astonishing discoveries of the 20th century. This is correct, but the real marvel is the development of antifungal resistance in hospitals, communities, and the environment concomitant with their use. Fungal infections have emerged as an important clinical threat, with significant associated morbidity and mortality. This study is desig...
متن کاملBiological Evaluation of Heterocycle Moiety of Some Novel azoles Derivatives as Antibacterial and Antifungal potential Agents
Background & Objective: Azole nucleuses are very important part of antimicrobial, analgesic and anti-inflammatory drugs. The azole class of compounds is the most popular among the antibacterial and antifungal classes because of its lower toxicity, higher efficacy and a broad spectrum of activity. Today, Efforts have focused on the development of new, less toxic and more efficacious antifungal a...
متن کاملIdentification of novel genes conferring altered azole susceptibility in Aspergillus fumigatus.
Azoles are currently the mainstay of antifungal treatment both in agricultural and in clinical settings. Although the target site of azole action is well studied, the basis of azole resistance and the ultimate mode of action of the drug in fungi are poorly understood. To gain a deeper insight into these aspects of azole action, restriction-mediated plasmid integration (REMI) was used to create ...
متن کاملAzole Resistance in Dermatophytes: Prevalence and Mechanism of Action.
Azole antifungal agents (eg, fluconazole and itraconazole) have been widely used to treat superficial fungal infections caused by dermatophytes and, unlike the allylamines (such as terbinafine and naftifine), have been associated with resistance development. Although many published manuscripts describe resistance to azoles among yeast and molds, reports describing resistance of dermatophytes ar...
